利用中藥提取殘渣培養熱帶靈芝的研究

English Abstract

Purposes: To explore the feasibility of utilizing extracted residues of Huangqi (Astragalus membranaceus) and Sanqi (Panax notoginseng) as culture media to cultivate high quality Ganoderma tropicum. Methods: The effects of the water-soluble components extracted from the herbal residues on the mycelium growth of G.tropicum, as well as on the accumulation of polysaccharides and triterpenoids were studied. The effects of Huangqi and Sanqi extracted residues on their fruiting body growth, as well as on the yields of polysaccharides and triterpenoids were also investigated. Polysaccharides were prepared by hot water extraction followed by ethanol precipitation process and determined by phenol-sulfuric acid method. Triterpenoids were prepared by ultrasonic extraction with methanol followed by ethyl extraction process and determined by high performance thin layer chromatography (HPTLC) system. Results: Huangqi and Sanqi residues could significantly promote the mycelium growth of G.tropicum. The mycelium yields were improved by 31% and 50%, respectively. The mycelium growth curves were drawn according to the dried mycelium weights. Appropriate harvest time was obtained according to the amount of polysaccharides. There was no triterpenoids detected in mycelium and culture broth. In addition, the Huangqi and Sanqi residues could also significantly promote the fruiting body formation, as well as the polysaccharides and triterpenoids yields. The yields of fruiting body were improved by 53% and 47% respectively. Furthermore, a relatively large amount of polysaccharides was detected in the solid fermentation medium. Conclusions: It is feasible and promising to cultivate G. tropicum with the extracted residues from Huangqi and Sanqi.

研究目的:本實驗以熱帶靈芝(Ganoderma tropicum)作為研究菌種,採用中 藥提取殘渣,黃芪(Astragalus membranaceus)藥渣和三七(Panax notoginseng) 藥渣作為培養基質,探究黃芪藥渣和三七藥渣培養熱帶靈芝的可行性,進而實 現中藥提取殘渣再利用并期望得到優質的熱帶靈芝。 研究方法:通過向液體培養基中加入不同濃度的藥渣水溶性成分,探討藥渣對 熱帶靈芝菌絲生長,胞內、胞外多糖積累,胞內、胞外三萜積累的影響;並在 熱帶靈芝固體培養基中加入不同比例藥渣,進一步探究藥渣對熱帶靈芝子實體 產量、子實體多糖產量、子實體三萜產量的影響,并檢測了發酵菌質的多糖產 量。水提醇沉法製備粗多糖樣品、苯酚硫酸法檢測多糖含量,甲醇提取及乙酸 乙酯萃取製備三萜、HPTLC 系統檢測三萜含量。 研究結果:實驗結果表明黃芪藥渣和三七藥渣水溶性成分對熱帶靈芝菌絲積累 均具有促進作用,與對照組相比分別提高了 31%和 50%;探究得到添加藥渣水 溶性成分培養熱帶靈芝菌絲生長曲線,結合胞內和胞外多糖產量分析得出合適 的菌絲收集時間。然而,液體培養未能檢測到菌絲和發酵液中含有三萜成分。 另外,兩種藥渣對熱帶靈芝子實體的生長均具有促進作用,與對照組相比分別 提高了 53%和 47%;添加藥渣培養得到的子實體三萜和多糖產量也更高,並且 此培養方式還得到了較高含量的菌質多糖。 結論及意義:本研究表明黃芪藥渣和三七藥渣可以大部分、甚至完全替代常規 熱帶靈芝培養料中棉籽殼和木屑,實現中藥提取殘渣的回收利用。添加藥渣培 養得到的熱帶靈芝樣品三萜和多糖產量更高,更突顯了中藥渣栽培熱帶靈芝的 意義。 關鍵詞:熱帶靈芝;黃芪藥渣;三七藥渣;多糖;三萜
