三種莪術抗腫瘤作用的比較及其活性成分探討

Ezhu is an famous and commonly used traditional Chinese medicine. It has been reported that Ezhu possess anti-tumor activity. There are three species of Curcuma including Curcuma phaeocaulis Val., Curcuma kwangsiensis S. G. Lee et C. F. Liang and Curcuma wenyujin Y. H Chen et C. Ling. Used as Ezhu as recorded in "The Pharmacopoeia of People's Republic of China" 2005 edition. However, it was reported that there were great different differences of the chemical components among the three species of Ezhu. In the present thesis, the research is focus on the differences of anti-tumor effects among the three species of Ezhu and their active components. Firstly, the anti-tumor effect of three species of Curcuma were tested on the human Hepatocarcinoma cell line HepG2, human premyelocytic leukemia cell line HL-60 and human breast adenocarcinoma cell line MCF-7 were used in the study. The results showed that all the three species of Ezhu had anti-tumor effect, among of them, the effect of Curcuma phaeocaulis is the best, Curcuma kwangsiensis is the better, and Curcuma wenyujin showed the weakest of the three. However, the volatile oil from Curcuma wenyujin showed stronger anti-tumor effect than the volatile oil from Curcuma kwangsiensis. Then the IC50 of different species of Ezhu were coupled to the data of GC-MS and HPLC analyses results. The principal components method of factor analysis and Pearson of bivariate correlations analysis were performed by using SPSS for Windows 15.0. We hope the statistics analysis can provide research method to explore the active components of Ezhu, and provide a theoretical basis for the further development of Ezhu. As the result, the second factor of GC-MS analysis contains the peak of No. 40,51,54,56,58,72,76,85,91,96,98,100,107,108, and the third factor of HPLC analysis contains the peak of No.5,12,13. The peaks of No. 5 and No.54 are curcumenol, and the peak of No.13 is furanodienone. The second factor of GC-MS analysis and the third factor of HPLC analysis have great variance contribution rate, so they may have prominent correlation with the anti-tumor effect. They may contribute to anti-tumor effect as a group of active component, and further study is needed in the future. 澳門大學碩士學位論文 中文摘要 莪術是常見的活血化瘀的中藥,臨床應用廣泛,研究表明其在抗腫瘤方面療 效顯著。《中華人民共和國藥典》2005年版一部共收錄了3種 術,即蓬 術( 術) Curcuma phaeocaulis Val,廣西莪術(桂術) Curcuma kwangsiensis S. G. Lee et C. F. Liang 和溫郁金(溫莪術) Curcuma wenyujin Y. H Chen et C. Ling的根莖 均作為莪術入藥。 雖然蓬莪術、廣西莪術和溫莪術均作中藥術使用,但是研究發現三種我術 的化學成分存在較大的差異。因此,本實驗對不同來源的莪術抗腫瘤的作用的進 行了比較研究,對化學成分的差異和抗腫瘤的作用的差異的相關性進行了探討。 首先考察了三種莪術對不同腫瘤細胞株的作用,結果表明,三種莪術對體外 培養的人急性早幼粒細胞性白血病細胞株HL-60有較強的抑制作用,其次對人乳 腺癌細胞株MCF-7也表現為抑制細胞增殖的作用,對人肝癌細胞株HepG2的抑制 增殖的作用稍差。並且義術對這些腫瘤細胞的抑制作用呈劑量依賴性。另外,不 同莪術的抗腫瘤作用效果具有差異性,其中蓬莪術效果最好,其次為廣西莪術, 再次為溫 術,但廣西装術油的效果差於溫莪術油。 然後將義術抗腫瘤的藥理數據IC5與GC-MS和HPLC分析的色譜數據相結 合,利用SPSS for Windows 15.0中因數分析的主成分分析法和Pearson相關分析 法,對我術的化學成分進行統計分析,為確定義術有效的抗腫瘤物質基礎提供研 究方法,並為進一步開發義術提供理論依據。分析結果提示GC-MS色譜峰因數 分析後第二因數中序列號為40,51,54, 56,58,72,76,85,91,96,98, 100,107,108的色譜峰所代表的化合物,和HPLC色譜分析後第三因數中的序列 號為5,12和13號色譜峰所代表的化合物可能是我術的抗腫瘤的物質基礎;其中5 號和154號峰均為莪術烯醇,13號峰為味喃二烯酮。第二因數HPLC色譜,可能作 為有效成分組,成為莪術的抗腫瘤的活性成分。 關鍵字: 術,抗腫瘤,色譜分析,SPSS for Windows 15.0,主成分分析,活性 成分
