Study on anthraquinones from morinda officinalis

English Abstract

Morinda officinalis (family Rubiaceae), is a traditional Chinese tonic herb mainly distributed in Guangdong, Guangxi and Fujian provinces in China. It has been extensively used for tonifing kidney, dispelling wetness, muscular and skeletal atrophy in China for about two thousand years. It mainly treat lack of libido, impotence, infertility, painful knees and legs, arthritis, fatigue, musculoskeletal atrophy, anti-rheumatic. The modern pharmacology research shown it also possesses anti-bacterial, antiosteoporotic activity. Although the chemical constituent of Morinda officinalis had been studied by some researchers ten years ago, in order to analyze and explore its constituents further, and hope it could provide good guarantee to quality control, we continue to concentrate on its chemical constituent. This thesis main covers chemical constituent separation process and seven anthraquinone quantitative analysis. The thesis contained four chapters: Chanter l. Review of the chemical constituents pharmacology and the quality contes studies of Morinda officinalis. Chapter I: Isolation of chemical constituent from Morinda oficinalis. richiten compounds had been isolated from this herb by various cromatographic methods. The molecular structures of these compounds were elucidated by NMR spectrum and chemical methods as follows: 1-hydroxy 2-methyanthraquinone (1) physcion (2), digiferruginol (3). lucidin co-ethyl ether (4), 2-hydroxy-I-methoxyanthraquinone (S), rubiadin (6), rubiadin-1-methylether (7), 1,3-dihydroxy-2-hydroxymethyl-anthraquinones (8), anthraquinone-2-carboxylicacid (9), 1,3-dihydroxy-2-methoxyanthraquinone (10), 1,2-dimethoxy-3- hydroxyanthraquinone (11), alizarin (12), 3-hydroxy-2-methyl. anthracene-9, 10-dione (13), stigmasterol (14), B-sitosterol (15), 7-hydroxy-6-methoxy-coumarin (16), daucoterol (17) and fumaric acid (18). 2. Compound 3, 8, 9, 12, 13, 14, 17 and 18 were isolated from this herb for the first time. Chapter III: Quantitative analysis of seven anthraquinones in Morinda officinalis by using pressurized liquid extraction and high performance liquid chromatography. Chapter IV: Conclusion and significance of the project.

巴載天是茜草科植物巴戟天的乾燥根,主要分佈廣東、廣西、福建,四/1 等地。巴戟天在中國使用有兩千年之久,具有補腎陽,強筋骨,祛風濕之功效,王要用於治療陽痿遺精,宮冷不孕,月經不調,少腹冷痛,風濕痹痛,筋骨痠軟。現代藥理學證明巴戟天中的某些成分具有抗菌和抗骨質疏鬆等活性。之 前,有人也對巴戟天的化學成分進行了研究,並取得了一定成績。為了進一步研 究和闡明其化學組成並希望為其之後品質控制提供物質保證。因此,本論文針對 巴戟天的化學成分及其七個蔥醌成分的定量分析方法展開了研究。 全文可分為四章 第一章是有關巴戟天的文獻綜述,包括巴戟天的化學成分和藥理學研究,以及巴 戟天的品質控制的研究。 第二章巴戟天的主要化學成分的分離。 1.應用多種色譜方法從巴戟天中分離了 18 個化合物,並通過光譜學和化學 的方法鑒定他們的結構為: 1-羥基-2-甲基蔥醌(1),大黃素甲醚(2) Digiferruginol (3), 1,3一二 羥基一乙氧甲基一蔥醌(4),2-羥基-1-甲氧基蔥醌(5),甲基異茜草素(6),甲基異茜草素 -1-甲醚(7),1,3-二羥基-2-甲氧基蔥醌(8),蔥醌 -2-羧酸(9),1,3-二甲氧基-2-羥基蔥醌(10),1,2-二甲氧基-3-羥基蔥醌 (11),茜素(12),3-輕基-2-甲氧基蔥醌 (13),豆甾醇 (14),穀甾醇 (15),7-經基-6-甲氧基-香豆素(16),8殺街醉葡萄糖苷 (17)和 丁烯二酸 (18) • 2.化合物3,8,9,12,13,14,17 和 18為首次從該植物中分離。 第三章加壓溶劑提取-高效液相色譜法同時測定七個蔥醌成分。 第四章 總結。 關鍵宇:巴戟天;蔥醌;香豆素;HPLC
