Abstract In order to search for the bioactive compounds from a tradional Chinese medicine Pueraria lobata, systematic chemical study on P. lobata were carried out. Gegen is the dried root of Pueraria lobata (Willd.) Ohwi. It tastes sweet, spicy and cool. It has effects on ptomoting saliva, penetrating exanthema and inhabiting diarrhea. Puerarin, isoflavnoids regarded as one of the main ingredients in this plant, is not only the special ingredient but also the main effective ingredient which has been applied on curing for cardiovascular diseases extensively. In order to search for new active compounds, we commited systematic chemical study on P. lobata. 42 compounds have been isolated from the 95% EtOH extract of this plant by using several chromatographic separation techniques and purification methods. Meanwhile, 32 compounds of them were identified according to their physico-chemical and spectral data as follows: dadzein (GG-1), formononetin (GG-2), 3'-methoxy daidzein (GG-3), dadzin (GG-4), genistin (GG-5), ononin (GG-6), 8-methoxy ononin (GG-7), sissotorin (GG-8), daidzein4', 7-diglucoside (GG-9), puerarin (GG-10), 4'-methoxypuerarin (GG-11), 3'-methoxy puerarin (GG-12), 3'-hydroxypuerarin (GG-13), daidzein-8-C-apiosyl(1→6)glucoside (GG-14), 6"-O-B-D-xylosypuerarin (GG-15), 3'-hydroxy-daidzein-8-C-apiosyl(1-6)glucoside (GG-16), 3'-hydroxy-6"-O-B-D-xylosyl puerarin (GG-17), 3'-methoxy-6"-O-B-Dglucosylpuerarin (GG-18), 3'-hydroxy-formononetin-8-C-apiosy-(1→6)glucoside (GG-19), Puerarin-4'-O-gluoside (GG-20), 3'-hydroxy-4'-O-glucosylpuerarin (GG-21), 3'-methoxy-4'-O-glucosylpuerarin (GG-22), mirificin-4'-O-B-D-glucoside (GG-23), sophoracoumestan A (GG-24), allantion (GG-25), puerarol (GG-26), B-sitosterol (GG-27), lupeol (GG-28), lupeone (GG-29), (6R, 7E,9R)-3-oxo-a-ionol-9-O-f-Dglucopyranoside (GG-30), sucrose (GG-31), (-)-puerol B 2-0-glucopyranoside (GG-32). Four of them, GG-17, GG-18, GG-19 and GG-22, are new compounds. Bseides, GG-7, GG-28, GG-30 and GG-31 are separated from this genus for first time and GG-24, GG-29 are separated from this species for first time. 摘要 為尋找傳統中藥葛根生物活性成分以及為其品質控制和藥理評價提供物質基 礎,我們對中藥葛根的化學成分進行了系統研究。 葛根為豆科植物野葛 Pueraria lobata (Wild.) Ohwi 的乾燥根,習稱野葛。其味 甘辛涼,歸脾胃經,具有生津,透疹,升陽止瀉之功效。異黃酮類化合物是葛根的 主要成分;其中葛根素是葛根的特有成分,在治療心血管系統疾病上有著廣泛的 應用。為了尋找新的活性化合物,本課題對葛根的化學成分進行了系統研究,運 用多種色譜分離和純化手段,從葛根95%乙醇提取物中分離得到42個化合物並根 據理化和波譜資料鑒定出32 個結構,分別為大豆 元(GG-1),芒柄花素(GG-2), 3'-甲氧基大豆苷元(GG-3),大豆 (GG-4),染料木苷(GG-5),芒柄花苷(GG-6),8- 甲氧基芒柄花苷(GG-7),5-羟基芒柄花苷(GG-8),大豆苷元-4', 7-二葡萄糖苷(GG9),葛根素(GG-10),4-甲氧基葛根素(GG-11),3-甲氧基葛根素(GG-12),3'-羟基 葛根素(GG-13),大豆苷元 8-C-芹糖(1→6)葡萄糖苷(GG-14),葛根素木糖苷 (GG-15),3'-羟基大豆 元8-C-芹糖(16) 葡萄糖苷(GG-16),3'-羟基葛根素木糖苷 (GG-17),3'-甲氧基葛根素葡萄糖苷(GG-18),3-基芒柄花素8-C-芹糖(16)葡萄 糖苷(GG-19),葛根素-4'-O-葡萄糖苷(GG-20),3-羟基-4'-0-葡萄糖葛根素(GG-21), 3-甲氧基-4-O-葡萄糖葛根素(GG-22),美佛辛-4'-O-葡萄糖苷(GG-23), 香豆素 A (GG-24), 囊素(GG-25),葛香豆雌酚(GG-26),β-穀醇(GG-27),羽扇豆酮(GG-2 8),羽扇豆醇(GG-29), (6R, 7E, 9R)-3-oxo-a-ionol-9-0-B-D-glucopyranoside (GG-30),蔗糖(GG-31), (-)-Puerol B 2-O-glucopyranoside (GG-32)。其中 GG-17, GG-18,GG-19,GG-22 為新化合物,GG-7,GG-28,GG-30,GG-31 為從本屬植 物中首次分離得到,GG-24,GG-29 為從本種植物中首次分離得到。
