Quality evaluation of Salvia miltiorrhiza based on multiple components determined by twice development thin layer chromatography

Danshen, the dried root and rhizome of Salvia miltiorrhiza, has been frequently used to treat various ailments, especially cardiovascular diseases. Recent researches have revealed Danshen's possible benefits in ischemia or coronary artery diseases, renal diseases, alcohol temperance, liver diseases and so on, which were contributed by both water-soluble salvianolic acids and fat-soluble tanshinones in S. miltiorrhiza. According to the Chinese Pharmacopoeia (2005), the identification of Danshen is performed by thin layer chromatography (TLC) on tanshinone IIA and salvianolic acid B, while the quantification of this herb is done by high performance liquid chromatography (HPLC) on the aforementioned compounds. Recent researches have revealed many improved HPLC methods of determining tanshinones and salvianolic acids for the quality assessment of Danshen. In fact, TLC is a quick and convenient way for qualitative and quantitative analysis, but former studies have only analyzed the hydrophilic and lipophilic bioactive constituents in Danshen on separate TLC plates. None of them have analyzed these two groups of compounds simultaneously on one single plate. In the present research, a twice development TLC method has been established for simultaneous qualitative and quantitative determination of twelve hydrophilic and lipophilic constituents, namely sodium danshensu, salvianolic acid B, rosmarinic acid, salvianolic acid A, caffeic acid, protocatechuic acid, protocatechuic aldehyde, dihydrotanshinone I, cryptotanshinone, tanshinone I, tanshinone IIA and miltirone, in Salvia miltiorrhiza. Optimized developing solvents dichloromethane-ethyl acetate-formic acid (22:24:10)and petroleum ether-ethyl acetate-cyclohexane (25:11:14) were used for the twice development on silica gel 60F254 plates. Their characteristic TLC profiles were observed under UV 254 nm and 365 nm, while the quantification was achieved by scanning at 260 nm or 290 nm with reference wavelength of 400 nm. In addition, fingerprints and hierarchial cluster analysis were also performed based on the TLC profiles and quantitative results, which is helpful for Danshen's quality evaluation. 丹參爲唇形科植物丹參 Salvia miltiorrhiza Bge.的乾燥根和根莖,可用於治療心肌缺血或冠心病、腎病、肝病以及戒酒等。其活性成份主要水溶性的酚酸和脂溶性的丹參酮類成份。 根據中國藥典2005年版的記載,丹參的鑑別是用薄層色谱法對丹參酮IIA及丹酚酸B分別進行檢测,而其定量方法則是以高效液相色譜法测定上述化合物。近年的丹參質量評價方法研究主要針對高效液相色譜法,對丹參酮類和丹酚酸類進行分析。實際上,薄層色譜法是一種快捷方便的定性定量分析方法,此前的研究多把丹參的水溶性和脂溶性成份在不同的薄層板上进行分析,而未能在同一张薄層板上同時對這兩類化合物進行分析。 本研究建立了二次展開薄層色譜掃描法同時對丹參的十二種水溶性和脂溶性成份丹參素鈉、丹酚酸 B、迷迭香酸、丹酚酸A、咖啡酸、原兒茶酸、原兒茶醛、二氫丹參酮I、隱丹參酮、丹參酮I、丹參酮 IIA 和丹參新酮進行定性定量分析。在納米—硅膠 60F254薄層板上使用經過優化的展開劑二氨甲烷:乙酸乙酯:甲酸(22:24:10)以及石油醚:乙酸乙酯:環己烷(25:11:14)進行二次展開。在紫外254 nm 和 365 nm 下拍照記錄其特徵薄層圖譜,並以 400 nm 爲參比波長,在260 nm 或 290 nm 下進行薄層掃描及含量測定。此外,基於薄層圖譜和定量結果進行了指紋圖譜和聚類分析,有助於丹參的品質評價。 關鍵詞:丹參,酚酸,丹參酮,薄層色譜法,二次展開,聚類分析,指紋圖譜
